Optically Computed Phase Microscopy for Functional Imaging of Label-Free Cells

Abstract

It is crucial to understand the dynamics of cells when they interact with one another and interact with their environment. For example, mRNA Covid vaccines are delivered into the cells via lipid nanoparticles. Imaging the mechanical interactions between cells and nanoparticles is critical to advance the efficiency and safety of these drug delivery systems. To image the mechanical dynamics of label-free cells, our team investigate a novel imaging technology, optically computed phase microscopy (OCPM) that processes 3D spatial-spectral data through optical computation, addresses 3D spatial locations using 2D photon detection, achieves sub-micrometer spatial resolution, and detects motion with nanometer sensitivity. We demonstrated OCPM imaging in 3D surface profile reconstruction of label-free cells, quantitative observation of the cell detachment process, and Doppler imaging of cell-particle interaction. We envision OCPM to have a wide range of applications in various fields of biomedicine.

Biography

Dr. Xuan Liu received her bachelor’s degree in Electrical Engineering from Tsinghua University and her Ph.D. in Electrical Engineering from the Johns Hopkins University. She is currently an Associate Professor in the Department of Electrical and Computer Engineering, at New Jersey Institute of Technology. Dr. Liu’s expertise lies in the instrumentation and signal processing of biomedical optical imaging. Her research focuses on optical coherence tomography for biomedical applications, and functional phase microscopy for dynamic cellular imaging. The research at Liu’s lab has been supported by National Cancer Institutes, National Institute of General Medical Sciences, Nokia Bell Labs, and other agencies.
